Currently, it is known that the transduction of Hh signaling is regulated through Ptch1-Smo-Gli1 [ 6, 7, 8]. In the absence of Hh ligands, the twelve-transmembrane receptor Patched1 (Ptch1) suppresses the activity of seven-transmembrane receptor Smoothened (Smo), which is homologous to the G-protein coupled receptors, and leads to negatively regulate the Hh signaling pathway. However, in the presence of Hh ligands, Hh ligands directly bind to Ptch1 and alleviate its inhibition of Smo [ 9, 10, 11]. As a consequence, the activated Smo promotes Gli transcription factors to translocate into nucleus and regulates target genes expression. Measurements were performed using a Prometheus NT.48 instrument (NanoTemper Inc., Germany). ShhN was diluted to 0.2 mg/mL in buffer contained 10 mM HEPES, pH 7.4, 150 mM NaCl, and 5% DMSO. Measurements were carried out using 70% power with standard Prometheus NT.48 capillaries. The heating was 1.0°C/min from 20 to 95°C. Results and Discussion Discovery of ShhN–Ptch PPI Inhibitors Compared to the reported small-molecular-weight ShhN inhibitors, the IC 50 of compound 7_3d3 is about 10 times stronger than robotnikinin (IC 50 measured using the same assay in the present study); HL2-m5, a macrocycle peptide with a molecular weight of 1633 and an IC 50 of 0.25 μM (determined using a similar cell-based assay as robotnikinin), 7_3d3 is much smaller with a molecular weight <500, and the non-cyclic structure makes it more suitable for further optimization.
