The typical detection methods of trace metal and metalloid elements in serum include chemical analysis, spectrophotometry, atomic absorption spectrometry, and atomic fluorescence spectrometry [ 14, 15]. The most significant disadvantage of these methods is that they can only analyze a single element, and the sample pretreatment is complicated. Inductively coupled plasma mass spectrometry (ICP-MS) is a relatively new detection technology that is widely used to analyze environmental, food, and biological samples. This method has the advantages of high speed, wide linear range, high precision, good accuracy, low detection limit, and low sample quantity requirements. It can simultaneously determine multiple elements and is suitable for testing large quantities of samples [ 16, 17].

The pre-established acceptance criteria for a multiplexed method is CV < 25% for inter-assay precision and < 20% for intra-assay precision (based on common published parameters 18, 19, 29). All cytokines met acceptance criteria of CV < 20% for intra-assay precision (Fig. 1C, Supplemental Table 7). Inter-assay CV was acceptable (< 25%) for 22 of 23 cytokines; TNFα showed an unacceptable inter-assay CV of 45.3% (Fig. 1C, Supplemental Table 7). As expected, inter-assay CVs were generally higher than intra-assay CVs, demonstrating the day-to-day variability of a technically challenging assay such as this one. In comparison to the manufacturer-reported intra-assay precision, the current study agreed with the manufacturer reported CV of < 5% in 18 of 23 (78%) analytes for intra-assay precision and the manufacturer-reported CV of < 15% in 20 of 23 (87%) analytes for inter-assay precision (Fig. 1C). By comparing serum and plasma from multiple donors, it becomes clear that a simple “conversion factor” between serum and plasma cannot be derived, even on an individual cytokine basis. This is because the ratio of serum to plasma MFI varies significantly between donors ( Fig. 1b), at least for many of the cytokines tested. We also show, through spike recovery experiments, that plasma and serum both have significant inhibitory effects, that are variable between individuals, and that are only partially compensated for by the manufacturer’s standard dilution buffer ( Figs. 3, ​ ,4, 4, ​ ,5). 5). However, serum appears to have a slightly greater inhibitory effect for some cytokines. Interestingly, this matrix effect is not seen, or seen to a much lesser extent, in the MSD assay compared with either magnetic or polystyrene bead Luminex assays ( Fig. 4).
